 For crabs and other crustaceans animals which couldn’t secret bile acids and convert into cholesterol(Kanazawa and Koshio, 1994). As one necessary nutrition, steroid is important for their physiological function and normal growth, which are the base materials for cell and molting hormone.There is experiment (from sheen 1994) shows that the penaeus monodon would show lower survival rate and have effects on growth speed without cholesterol in diet.what’s more, cholesterol also can improve the anti-stress ability for low salinity (Paibulkichakul 1998). Adding bile acids in diet of penaeus monodon can improve the utilization of cholesterol by the function of completely emulsified. The experiment aims to study the effects of growth performance and blood biochemical index of penaeus monodon by using bile acids to replace part of cholesterol in diet, which can provide the evidence for saving feed cost.

1.    Material and methods

1.1 Experiment Diets

Control group fed with complete feed make up by the main materials like: peru fish meal, soybean meal ,flour and 0.1% cholesterol , while experiment groups respectively adding 100 mg/kg ,200 mg/kg ,300 mg/kg ,400 mg/kg and 500 mg/kg bile acids ( >95% content provide by shandong longchang animal health product CO., Ltd ) to replace 10% , 20%,30%, 40% and 50% cholesterol , and named as Experiment group 1,2,3,4,5 respectively. All the diets were made as pellets (<1.0 mm), conserve under -20℃condition.

1.2 Experiment shrimps and feeding management

All the experiment penaeus monodons from Nanhai aquaculture research laboratory, department of China Aquaculture Research Laboratory. 540 tails larval penaeus monodon (30 days old) are divided into 6 groups, 3 duplicates per group, and 30 tails shrimp per duplicate. Every duplicate put in glass tank (0.5 m3) , filtered sea water used as feeding water , with oxygenator and change water each day, keep 28-30℃water temperature and 30.0-32.0 salinity. Three times Feed intake at 8::00、15:00 and 20:00 respectively. The experiment process with 30 days.

1.3  Measurement of growth performance

Weight at the end of the experiment, the indexes are figured out as follows:

Weight gain rate (WGR) /% =(final weight -initial weight)/initial weightx100;

FCR=feed intake /total weight gain;

Survival rate /%=shrimp number at the end /shrimp NO at the beginningx100

1.4  Measurement of blood biochemical index

At the end of the experiment, blood sample are draw from cavum pericaradial of shrimp, and put into centrifuge tube, kept under 4℃. After solidification, blood serum are separated and test the content of Triglyceride, cholesterol, glutamic pyruvic transaminase (SGPT) and glutamic oxalacetic transaminase (SGOT).

1.5  Analysis

Data for all response variable were analyzed one -way designs by analysis of variance using the SPSS 13.0 procedure of ANOVA system and Duncan’s multiple range test, which were used to determine treatment differences. All statement of significance is based on the probability level of 0.05.

2. Experiment results

The growth performance of every group showed on table 1. From the results ,we can see that adding bile acids can significantly improve growth performance of penaeus monodon, however , as the replacement of cholesterol increase, the growth rate become slow.Experiment group 1 shows that replace 10% cholesterol with the best WGR comparing with other groups (P<0.05), WGR on experiment group 2 and experiment group 3 have no big difference compared with control group. however, experiment group 4 and 5 shows a little snow WGR comparing with control group. 

For the FCR, compared with control group, experiment group 1 shows that adding bile acids to replace 10% with the best FCR data (P<0.05), experiment group 2 and experiment group 3 shows that there are no big difference to replace 20% and 30% cholesterol by adding bile acids , while adding bile acids to replace 40% and 50% cholesterol slightly higher on FCR.

What’s more , data on experiment 1, 2 ,3 shows that adding bile acids to replace 10%--30% cholesterol has no effects on survival rate compared with control group, while there are low survival rate when replace 40%-50% cholesterol on experiment group 4 and experiment group 5(P<0.05).

From the data we can see that compared with control group ,experiment group 1 (replacing 10% cholesterol )with the highest triglyceride content , and the content decrease when replace more and more cholesterol, while experiment group 4 and experiment 5 with the lowest content(P<0.05)

For the content of cholesterol , compared with control group , experiment group 1 with the lowest cholesterol content(P<0.05), and the content of cholesterol in blood increase while using bile acids to replacing more and more cholesterol in diet, the experiment group 4 and 5 with the highest blood content of cholesterol , which are higher than control group and experiment group 1 and experiment group 2 (P<0.05).

Content of SGPT in blood sample increase while replacing more cholesterol , experiment group 4 and experiment 5 with higher SGPT content compared with other groups(P<0.05). While , experiment group 1 has the lowest SGOT content when using bile acids to replace 10% cholesterol(P<0.05), experiment group 4 ,5 with the highest SGOT content , there are no big difference for other groups .

3. Results

There are no significant differences on growth performance and blood biochemical index for penaeus monodon when using bile acids to replace 10%-20% cholesterol in dietary.
